Properties
Domperidone is a dopamine antagonist with antiemetic properties. Domperidone does not penetrate well through the BBB. The use of domperidone is rarely accompanied by extrapyramidal side effects, especially in adults, but domperidone stimulates the release of prolactin from the pituitary gland. Its antiemetic effect may be due to a combination of peripheral (gastrokinetic) action and antagonism to dopamine receptors in the chemoreceptor trigger zone, which is outside the BBB.
When administered orally, domperidone increases the duration of antral and duodenal contractions, accelerates gastric emptying, and increases lower esophageal sphincter pressure. Domperidone has no effect on gastric secretion.
Indications
— to relieve symptoms of nausea and vomiting.
Dosage and administration
The drug is taken orally. It is recommended to take Motil Fast sachets 15-30 minutes before a meal, if the drug is taken after a meal, the absorption of domperidone may slow down.
Adults and adolescents over 12 years of age and children weighing ≥35 kg — 1 sachet. (10 mg) 3 times / day, the maximum daily dose is 3 sachets. (30 mg).
Children under 12 years of age and weighing 35 kg or more — 1 sachet. (10 mg) 3 times / day, the maximum daily dose is 3 sachets. (30 mg).
Contraindications
— hypersensitivity to domperidone or any other component of the drug;
-prolactinoma;
— simultaneous use of oral forms of ketoconazole, erythromycin or other potent inhibitors of the CYP3A4 isoenzyme, causing a prolongation of the QT interval, with the exception of apomorphine;
Side effects
Mental disorders: infrequently — irritability, nervousness.
From the nervous system: often — dizziness; rarely — convulsions; frequency unknown — extrapyramidal disorders.
From the side of the cardiovascular system: the frequency is unknown — ventricular arrhythmia *, ventricular tachycardia of the «pirouette» type, sudden coronary death *.
